Description
Protein A has been reported to bind strongly to rabbit IgG and mouse IgG2 isotypes. However, protein A does not bind mouse IgM and weakly binds IgG1. When conjugated to agarose, protein A acts as a solid matrix facilitating the immunoprecipitation of antigen-antibody complexes. It can also be utilized for antibody purification. As a horseradish peroxidase conjugate, protein A can be used instead of secondary antibody detection methods in Western blotting with polyclonal antibodies and with IgG2a and IgG2b mouse monoclonal antibodies.
Horseradish peroxidase (HRP) was coupled to protein A at a concentration of 4 mg/mg of protein A using the periodate method. For the detection of antigen using polyclonal antibodies in Western blotting, a concentration of 0.5 µg/ml (1:2000 dilution) is suggested.
